Asensitivereagentformeasuringthefreesulfhydrylcontentinproteins,peptides,andtissues.Usedtocharacterizereactivethiolgroupsandphotometricdeterminationofthiolsandformeasuringlow-molecularmassthiolssuchasglutathioneinbothpuresolutionsandBIOLOGicalsamples,suchasblood.Itcanalsomeasurethenumberofthiolgroupsonproteins.Throughreactionwithaliphaticthiolgroupsamixeddisulfideofproteinthiolandonemoleof2-nitro-5-thiobenzoatepermoleofproteinsulfhydrylgroupisbeingformed.DTNBhaslittleabsorbance.Reactionwith-SHgroupsonproteins(fromanysolventaccessIBLeCys)undermildalkalineconditions(pH7-8)producesthe2-nitro-5-thiobenzoateanion,whichgivesanintenseyellowcolorwithanabsorptionmaximumat409.5nm(Extinctioncoefficient:14150M-1*cm-1).Sensitivetovariousbufferions,therefore,theextinctioncoefficientusedtocalculatethenumberofsulfhydrylgroupsmustbematchedtothereactionconditions.Incasethethiolgroupsareindisulfidebonds,theymustbereducedunderanaerobicconditionspriortoreactionwithDTNB.
