The2,3-diaminonaphthalene(DAN)assayisroutinelyusedinthedeterminationofnitrite/nitratelevelsinBIOLOGicalfluidsandcellularextractsasoneindicatorofnitricoxideactivity.Theassay,asreportedbyMisko,usesthereactionofDANwithNO2-underacidicconditionstoformadetectablefluorescentnaphthotriazole.ThereactionproceedsatacidicpHatroomtemperature.FluorescenceismonitoredfollowingtheadditionofNaOH,whichraisespH,resultinginlowerbackgroundandincreasedsensitivity(Ex/Em:~365/415nm).However,detectionat450nmisrecommendedtoavoidfluorescentblanksandincreasesensitivity.ThefluorescentbackgroundofDANislowformaximumsensitivity.DetectionlimitsofNOachievedbythismethodareinthehigh-pMrange.2,3-Diaminonaphtaline(DAN)isusedasaderivatisation-reagentforthedeterminationofseleniumatlowdetectionlimits.The4,5-benzopiaselenol-complexcanbequantifiedbyphotometryorGC.
