D-(+)-Biotin,alsoknownasvitaminHorcoenzymeR,isawater-solubleB-vitamin(vitaminB7).D-(+)-BiotinisacofactorresponsIBLeforcarbondioxidetransferinfivecarboxylaseenzymes,suchasacetyl-CoAcarboxylaseα,acetyl-CoAcarboxylaseβ,methylcrotonyl-CoAcarboxylase,propionyl-CoAcarboxylaseorpyruvatecarboxylase.Itisimportantinfattyacidsynthesis,branched-chainaminoacidcatabolismandgluconeogenesis.Itcovalentlyattachestotheepsilon-aminogroupofspecificlysineresiduesinthesecarboxylases.ThisbiotinylationreactionrequiresATPandiscatalyzedbyholocarboxylasesynthetase.Biotinisalsocovalentlyattachedtodistinctlysineresiduesinhistones,affectingchromatinstructureandmediatinggeneregulation.Biotiniswidelyusedthroughoutthebiotechnologyindustrytoconjugateproteinsforbiochemicalassays.Biotin"ssmallsizemeanstheBIOLOGicalactivityoftheproteinwillmostlikelybeunaffected.Thisprocessiscalledbiotinylation.Becausebothstreptavidinandavidinbindbiotinwithhighaffinity(Kdof10-14mol/lto10-15mol/l)andspecificity,biotinylatedproteinsofinterestcanbeisolatedfromasamplebyexploitingthishighlystableinteraction.Thesampleisincubatedwithstreptavidin/avidinbeads,allowingcaptureofthebiotinylatedproteinofinterest.Theattachmentofbiotintovariouschemicalsitesisusedtostudyvariousprocesses,includingproteinlocalization,proteininteractions,DNAtranscriptionandreplication.
