Kinases,suchasLIMkinase1(LIMK1)andLIMkinase2(LIMK2),havebeenidentifiedasparticipatinginsignalpathwaysaffectingactindynamicsbydeactivatingcofilin.OverexpressionofLMK1hasbeenfoundininvasivebreastandprostatecancercelllinesandsuppressionofLIMK2expressionhasbeenfoundtolimitmigrationofhumanfibrosarcomacells.Thus,theinhibitionofLIMK1and/orLIMK2enzymeshavebeensuggestedastargetsfortreatingcancer,includingreductionorpreventionofmetastasis.BMS-5isapotentinhibitoroftheLIMkinase.IthasIC(50)valuesof7nMand8nMforLIMK1andLIMK2respectively.LIMK1/2activitywasmeasuredviaTCAprecipitationassayusingabiotinylatedADF(actindepolymizationfactor)astheproteinsubstrateandthekinasedomainsofLIMK1andLIMK2astheenzymesourcesinthepresenceof1µMATP.
