Biotinderivative.SubstrateofthehorserADIshperoxidaseenzymeandusedasareagenttoamplifyimmunohistochemicalsignals.ItisbasedontheHRP-catalyzeddepositionoftyramideconjugates(suchasbiotinyl-tyramide)onasolidphase.Subsequentreactionwithstreptavidinfluorophoreresultsinthelocalizationofthefluorophoreatthesiteoftyramidedeposition.Thisfluorescence-basedtyramidesignalamplification(TSA)hasbeenwidelyusedinimmunohistochemistry,immunoelectronmicroscopy,fluorescentinsituhybridization(FISH)andfluorescenceELISA.TheTSAmethodhasbeenreportedtoincreasethedetectionsensitivityupto100-foldascomparedwithconventionalavidin–biotinylatedenzymecomplexprocedures.Itcanbeusedtogetherwithbothchromogenicandfluorescencevisualizationmethods.ItcanbeaddedtoanyotherstandardIHCprotocolandreducestheuseofotherreagents;improvessignaltonoisebyreducingthetiterofotherreagentsintheassayprotocolandenablesmulti-targetdetectioninbothIHCand(F)ISHapplications.
