Widely-usedfluorescentindicatorforestimatingintracellularpH.Thismembrane-permeantcompoundismodifiedbyintracellularesterasestoproduceBCECF,apolarfluoresceinderivativethatisretainedbycells.IntracellularBCECFisstableandhasaneffluxhalf-lifeofgreaterthantwohours.IntracellularpHmeasurementsaremadebyratioimaging,whichinvolvesdeterminingthepH-dependentratioofemissionintensity(detectedat535nm)whenthedyeisexcitedat490nmvs.theemissionintensitywhenexcitedat440nm.Thisapproachisamenabletoeitherspectrofluorometryorflowcytometry.BCECF-AMestercanalsobeusedtoinvestigateintracellularchangesinotherions,includingpotassium.Spectralproperties:λex490nm;λem535nm;λex=505nm;λem=520nminDMSO.
