AV-412isapotentdualinhibitorofEGFRandErbB2tyrosinekinases,includingthemutantEGFR(L858R,T790M),whichisclinicallyresistanttotheEGFR-specifickinaseinhibitorserlotinibandgefitinib.InenzymeassaysassaythecompoundinhibitedtheEGFRvariantsandErbB2inthenanomolarrangewithover100-foldselectivitycomparedwithotherkinases,apartfromABLandFLT1,whichwerebothmoderatelysensitivetothecompound.Incells,AV-412inhibitedautophosphorylationofEGFRandErbB2withIC(50)of43and282nM,respectively.Italsoinhibitedepidermalgrowthfactor(EGF)-dependentcellproliferationwithanIC(50)of100nM.Moreover,AV-412abrogatedEGFRsignalinginthegefitinib-resistantH1975cellline,whichharborsadoublemutationofL858RandT790MinEGFR.Inanimalstudiesusingcancerxenograftmodels,AV-412(30mg/kg)demonstratedcompleteinhibitionoftumorgrowthoftheA431andBT-474celllines,whichoverexpressEGFRandErbB2,respectively.
