AT7519isapotentinhibitorofseveralCDKfamilymembers.AT7519showedpotentantiproliferativeactivity(40-940nM)inapanelofhumantumorcelllines,andthemechanismofactionwasshownheretobeconsistentwiththeinhibitionofCDK1andCDK2insolidtumorcelllines.AT7519causedcellcyclearrestfollowedbyapoptosisinhumantumorcellsandinhibitedtumorgrowthinhumantumorxenograftmodels.TumorregressionwasobservedfollowingtwicedailydosingofAT7519intheHCT116andHT29coloncancerxenograftmodels.AlsoithasbeenshownthattheBIOLOGicaleffectsarelinkedtoinhibitionofCDKsinvivoandthatAT7519inducestumorcellapoptosisinthesexenograftmodels.Moreover,AT7519hasanattractivebiologicalprofileandiswelltoleratedandeffectivemakingitamoreplausIBLecandidateforclinicaldevelopmentthanpreviouslyavailableCDKinhibitors.IninvitrokinaseassaysAT7519showednanomolarlevelsofactivityfrom<10 to="" 2400="" for="" cyclins;="" only="" one="" other="" non-cyclin="" related="" kinase="" was="" inhibited="" at="" levels="" below="" 10µm="" (gsk3β,="" 89nm),="" all="" others="" tested="" had="" ic(50)"s="" of="" greater="" than="">
