Thecompound(Akt-I-1)inhibitsonlyAkt1(IC(50)4.6µM)anddoesnotinhibitAKT2,orAKT3.ThecompoundisareversIBLeinhibitor,andexhibitsalinearmixed-typeinhibitionagainstATPandpeptidesubstrate.InadditiontoinhibitingkinaseactivityofAKT1isoform,AKT-I-1blockedthephosphorylationandactivationofAKT1byPDK1(phosphoinositide-dependentkinase1).Theinhibitorwasfoundtobecell-activeandtoblockphosphorylationofAktatThr308andSer473,reducethelevelsofactiveAktincells,blockthephosphorylationofknownAktsubstratesandpromoteTRAIL(tumour-necrosis-factor-relatedapoptosis-inducingligand)-inducedapoptosisinLNCapprostatecancercells.
