AminoanalogofthecommonD-luciferinsubstrateforuseinmeasuringfireflyluciferaseactivity.Thiscellpermeablereagentcanbeusedwhenconsideringdesignofproluminescentbioconjugates(peptidasesubstrates)ofluciferinforultrasensitiveluminescentassays.Inbioluminescentproteaseassayspeptidesoraminoacidsareconjugatedtotheluciferinanalog6-amino-D-luciferin.Thesesubstratesareusefulforultrasensitiveanalysisofavarietyofproteaseenzymesincludingvariouscaspasesandcathepsins,chymotrypsin,trypsin,elastase,kallikrein,thrombin,ficin,bromoalain,plasmin,papain,ficinandmanyothersorevenforuseinvivotracingofluciferasetransfectedcellsinlivingtissues.Theresultingbioluminescencespectrumofreactionof6-Amino-D-luciferinisverysimilartothatobservedforthenaturalsubstrateD-luciferin.Peptidaseassaysusingabioluminogenicsubstratecanbeeasilyadaptedto96-wellformatsforHTS-basedassays.Usingthisprinciple,itispossIBLetodesignanassaytodetectalmostanypeptidaseorproteaseactivityviabioluminescencebyusingasubstratewithasuitablepeptidesequence.Applicationsinpathogendetection,discoveryofproteaseinhibitors,probingcellphysiologyandassessingproteaseactivityinoncogenesisarepossibleatextraordinarysensitivity.
