- Non-fluorescentreporterproteinsubstrate.Thisfluorogenicβ-glucuronidasesubstratereleasesthehighlyfluorescentdye5-dodecanoylaminofluorescein(Spectraldata:&lamBDa;ex=490nm,λem=520nm)indirectcorrelationwithenzymeactivity.
- Thesubstratescanpenetratethecellmembraneunderphysiologicalconditionsandformsanon-toxicgreenfluorescentproductthatiswellretainedinviablecellsandwhichallowssortingandcloningofthecellsandmonitoringofcelldevelopmentinvitroandinvivo.
- Thecolorlessandlipophilicsubstrateentersthecellsbyembeddingintheouterlayerofthecellmembraneasaglycolipidandthentransferringtotheinnermembranebya"flip-flop"mechanism.Onceinsidethecell,thesubstrateiscleavedbyβ-glucuronidase,generatingthegreenfluorescentenzymatichydrolysisproduct5-dodecanoylaminofluorescein,whichisretainedinthecellmembranebecauseofitslipophilichydrocarbonchain,andallowinghistochemical,spectrophotometricandfluorometricmeasurementsofGUSgenefusionexpression.Thissubstratehasbeenusedinβ-glucuronidase(GUS)geneexpressionkits.
