- Non-fluorescentreporterproteinsubstrate.Thisfluorogenicβ-glucosidasesubstratereleasesthehighlyfluorescentdye5-dodecanoylaminofluorescein(Spectraldata:λex=490nm,λem=520nm)indirectcorrelationwithenzymeactivity.Canbeusedforanalysisofcellviability.
- Thisfluorescentsubstrateisformeasuringthepresenceandactivityofaglycosidicenzymeinsideacell. Thesubstratescanpenetratethecellmembraneunderphysiologicalconditionsandformsanon-toxicgreenfluorescentproductthatiswellretainedinviablecellsandwhichallowssortingandcloningofthecellsandmonitoringofcelldevelopmentinvitroandinvivo.
- Thecolorlessandlipophilicsubstrateentersthecellsbyembeddingintheouterlayerofthecellmembraneasaglycolipidandthentransferringtotheinnermembranebya"flip-flop"mechanism.Onceinsidethecell,thesubstrateiscleavedbyβ-glucosidase,generatingthegreenfluorescentenzymatichydrolysisproduct5-dodecanoylaminofluorescein,whichisretainedinthecellmembranebecauseofitslipophilichydrocarbonchain,andallowinghistochemical,spectrophotometricandfluorometricmeasurements.
