10-acetyl-3,7-dihydroxyphenoxazin(AmplexRed)isanon-fluorescent,highlysensitiveandstableprobeforH2O2.AmplexRedisanapplicableasafluorogenicsubstrateforperoxidase.InthepresenceofhorserADIshperoxidase(HRP),theAmplexRedreagentreactsina1:1stechiometrywithH2O2toproducehighlyfluorescentresorufin.BecauseH2O2isproducedinmanydifferentenzymaticreactions,theAmplexRedreagentallowsresearcherstodetecttheactivityofmanydifferentenzymes.10-acetyl-3,7-dihydroxyphenoxazinhasabsorptionandfluorescenceemissionmaximaofapproximately563nmand587nm,respectively,andbecauseitsextinctioncoefficientishigh(54,000cm-1M-1),theassaycanbeperformedeitherfluorometricallyorspectrophotometrically.
