Verapamilhydrochlorideisanα-adrenergicreceptor(α-AR)antagoNISTandcalciumchannelproteininhibitorthatblockstheL-typeCa2+channelsinsmoothandcardiacmusclecells.Verapamilisanantiarrhythmicagentandvasodilatorknowntoreducetherenalclearanceofdigoxinandinduceapoptosisinprimaryandmetastaticcolonadenocarcinomahumancelllinesinvitro.Ithasbeenobservedthatverapamilcaninducecurrentsbyitself,presumablybyactingonthepotassiumandchlorideleakage.VerapamilhasalsobeenusedasaninhibitorofdrugeffluxpumpproteinssuchasMdr(P-glycoprotein).VerapamilisasubstrateofCYP3A4andCYP2C6.ItisalsousedinfluorescentcellsortingforDNAcontent,asitblockseffluxofavarietyofDNA-bindingfluorophoressuchasHoechst33342.Recentresearchhasshownverapamiltobeaneffectivetreatmentfordiabetesinanimalmodels.VerapamilhelpstreatdiabetesbylimitingTXNIPexpression.
