Polo-likekinase1(Plk1)isaregulatorofthecellcyclethathasbeenimplicatedinthepathologyofmanycancers.BI-2536isapotentandselectivesmall-moleculeinhibitorofmammalianPlk1.IthasinhibitoryactivityatsubnanomolarconcentrationsandinhibitstumorgrowthinmultipletumorlineswithIC(50)valuesbelow1µM.BI-2536alsoshowsan>1,000-foldselectivityforPIK1versusalargepanelofotherkinases.BI-2536alsodemonstratedlowKdvaluesagainstsisterkinasesPLK2andPLK3.PreclinicalstudiesinhumancancercelllineshaveshownthatBI2536disruptsspindleassembly,resultinginmitoticarrestandinducingapoptosis.BI-2536iscurrentlyinclinicaltrialsagainstseveraltypesofsolidtumorcancers.
