TRAIL-R1,-R2,-R3and-R4arereceptorsforthecytotoxicligandTRAIL.WhensignalingthroughTRAIL-R1and-R2,theadaptermoleculeFADDrecruitscaspase-8totheactivatedreceptor.Theresultingdeath-inducingsignalingcomplex(DISC)performscaspase-8proteolyticactivationwhichinitiatesthesubsequentcascadeofcaspases(aspartate-specificcysteineproteases)mediatingapoptosis.TheypromotetheactivationofNF-κB.TRAIL-R3lacksacytoplasmicdeathdomain,andTRAIL-R4containsatruncateddeathdomain,andhencearenotcapableofinducingapoptosis.TheyprotectcellsagainstTRAILmediatedapoptosisbycompetingwithTRAIL-R1andR2forbindingtotheligand.
