Zinc-α-2-glycoprotein(ZAG),firstidentifiedinthe1960s,derivesitsnamefromitsprecipitationfromhumanplasmaupontheadditionofzincsalts.ZAGhassincebeenfoundinsecretoryepithelialcellsandinarangeofbodyfluids.ZAGisidenticaltoalipidmobilizingfactorisolatedfromtheurineofpatientswithcancercachexiaandstimulateslipolysisininvivoexperiments.DuetoitsexpressioninandsecretionfromADIpocytes,ZAGisconsideredanadipokine.RecentlytheclinicalsignificanceofZAGhasbeenclarified.ZAGexpressioninadipocytesisinverselyrelatedtofatmass,thusitisintimatelyinvolvedinthemaintenanceofbodyweightinmiceandhumans.EpidemiologicalstudieshaveuncoveredanassociationbetweenZAGandplasmacholesterol.Thenon-synonymoussinglenucleotidepolymorphismrs4215inZAGisassociatedwithplasmacholesterolandobesity.StructurallyZAGpossessesaclassImajorhistocompatibilitycomplex(MHC)proteinfold.Itisdistinctfromothermembersofthisproteinfamilyinthatitissoluble,ratherthanbeinganchoredtoplasmamembranesanditassociateswithprolactininducIBLeproteinratherthanβ2microglobulin.Similartopeptideantigen-presentingclassIMHCmolecules,ZAGpossessesanopenapicalgroovebetweenitsα1andα2domainhelices.
