Disruptionoftheprecisebalanceofpositiveandnegativemolecularregulatorsofbloodandlymphaticvesselgrowthcanleadtomyriaddiseases.Althoughdozensofnaturalinhibitorsofhemangiogenesishavebeenidentified,anendogenousselectiveinhibitoroflymphaticvesselgrowthhasnotbeenpreviouslydescribed.Asplicevariantofthegeneencodingvascularendothelialgrowthfactorreceptor-2(VEGFR-2)thatencodesasecretedformoftheprotein,designatedendogenoussolubleVEGFR-2(esVEGFR-2/KDR)hasbeendescribed.TheendogenoussolubleesKDRinhibitsdevelopmentalandreparativelymphangiogenesisbyblockingVEGF-Cfunction.Tissue-specificlossofesKDRinmiceinduced,atbirth,spontaneouslymphaticinvasionofthenormallyalymphaticcorneaandhyperplasiaofskinlymphaticswithoutaffectingbloodvasculature.AdmiNISTrationofesKDRinhibitedlymphangiogenesisbutnothemangiogenesisinducedbycornealsutureinjuryortransplantation,enhancedcornealallograftsurvivalandsuppressedlymphangiomacellularproliferation.NaturallyoccurringesKDRthusactsasamolecularuncouplerofbloodandlymphaticvessels;modulationofesKDRmighthavetherapeuticeffectsintreatinglymphaticvascularmalformations,transplantationrejectionand,potentially,tumorlymphangiogenesisandlymphedema.
