Voltage-dependentanionchannel(VDAC)proteinsareabundant,pore-formingproteinsbelongingtotheeukaryoticmitochondrialporins.Itwasdiscoveredinthemitochondrialoutermembrane.Italsoisexpressedintheplasmamembrane.AtleastthreedifferentVDACgeneshavebeenidentifiedinvertebrates.VDACproteinsareknowntoplayanessentialroleincellularmetabolismandintheearlystagesofapoptosis.Forexample,VDACcontitutesamajorpathwaybywhichmetabolitessuchasADP/ATP,succinateandcitrateareexchangedbetweenthecytosolandmitochondria.AndVDAC1intheplasmamembraneestablishesanovellevelofapoptosisregulationputativelyviaitsredoxactivity.Formsachannelthroughthemitochondrialoutermembraneandalsotheplasmamembrane.Thechannelattheoutermitochondrialmembraneallowsdiffusionofsmallhydrophilicmolecules;intheplasmamembraneitisinvolvedincellvolumeregulationandapoptosis.Itadoptsanopenconformationatloworzeromembranepotentialandaclosedconformationatpotentialsabove30-40mV.Theopenstatehasaweakanionselectivitywhereastheclosedstateiscation-selective.Mayparticipateintheformationofthepermeabilitytransitionporecomplex(PTPC)responsIBLeforthereleaseofmitochondrialproductsthattriggersapoptosis.
