VASP(vasodilator-stimulatedphosphoprotein)isaproline-richproteinsubstrateofcAMP-andcGMP-dependentproteinkinases.VASPisanactin-associatedproteininvolvedinarangeofprocessesdependentonCytoskeletonremodelingandcellpolaritysuchasaxonguidance,lamellipodialandfilopodialdynamics,plateletactivationandcellmigration.VASPpromotesactinfilamentelongation.Itprotectsthebarbedendofgrowingactinfilamentsagainstcappingandincreasestherateofactinpolymerizationinthepresenceofcappingproteins.VASPstimulatesactinfilamentelongationbypromotingthetransferofprofilin-boundactinmonomersontothebarbedendofgrowingactinfilamentsanditplaysaroleinactin-basedmobilityofListeriamonocytogenesinhostcells.Itregulatesactindynamicsinplateletsandplaysanimportantroleinregulatingplateletaggregation.VASPphosphorylationisusedtomonitortheeffectofso-calledantiplateletdrugsthatreduceplateletreactivityandareusedtopreventstentthrombosis,strokesandheartattacksinpatients.PhosphorylationofVASPatSer157causesamobilityshiftinSDSgelelectrophoresisfrom46to50kDa,whichhasbeenusedasaconvenientMarkertomonitorcyclicnucleotide-dependentproteinkinaseactivity.
