ThetwoubiquitinC-terminalhydrolase(UCH)enzymes,UCHL1andUCHL3,deubiquitinateubiquitin-proteinconjugatesandcontrolthecellularbalanceofubiquitin.UCHL1andUCHL3arebothsmallproteinsof~220aminoacidsthatsharemorethan40%aminoacidsequenceidentity.UCHL3isuniversallyexpressedinalltissues,whileUCHL1isexpressedexclusivelyinneuronaltissue,testisandovary.TheactivityofUCHL3ismorethan200foldhigherthanUCHL1whenafluorogenicubiquitinsubstrateisused.UCHL1associateswithmonoubiquitinandUCHL3bindstoNedd8,ubiquitin-likeprotein.UCHL1andUCHL3playaroleintheregulationofneuronaldevelopmentandspermatogenesis.UCHL1isinvolvedinthepathogenesisofParkinson’sdisease(PD)andAlzheimer’sdisease(AD).Down-regulationandextensiveoxidativemodificationofUCHL1havebeenobservedinthebrainsofADpatientsaswellasPDpatients.Ubiquitin-proteinhydrolaseinvolvedbothintheprocessingofubiquitinprecursorsandofubiquitinatedproteins.ThisenzymeisathiolproteasethatrecognizesandhydrolyzesapeptidebondattheC-terminalglycineofeitherubiquitinorNEDD8.
