Ubiquitincarboxy-terminalhdlL1hydrolase(UCHL1)isadeubiqutinatingenzyme.UbiquitinUCHL1,alsoknownasPGP9.5,isaproteinof223aminoacidsandoneofthemostabundantproteinsinthebrain(1-2%ofthetotalsolubleprotein).Althoughitwasoriginallycharacterizedasadeubiquitinatingenzymerecentstudiesindicatethatitalsofunctionsasaubiquitin(Ub)ligaseandamono-Ubstabilizer.Alargeamountofmono-UbistightlyassociatedwithUCHL1,inhibitingthedegradationofmono-Ubinthebrain.ThepreciseregulationofUCHL1isessentialforneuronstosurviveandtomaintaintheirproperfunction.UCHL1isinvolvedinthepathogenesisofParkinson"sdisease(PD)andAlzheimer"sdisease(AD).Down-regulationandextensiveoxidativemodificationofUCHL1havebeenobservedinthebrainsofADpatientsaswellasPDpatients.Apost-translationalmodificationofUCHL1thatcontrolsthefunctionofUCHL1ismono-ubiquitination.ItoccursreversIBLytoalysineresidueneartheactivesiteofUCHL1.Ubiquitin-proteinhydrolaseinvolvedbothintheprocessingofubiquitinprecursorsandofubiquitinatedproteins.ThisenzymeisathiolproteasethatrecognizesandhydrolyzesapeptidebondattheC-terminalglycineofubiquitin.Alsobindstofreemonoubiquitinandmaypreventitsdegradationinlysosomes.ThehomodimermayhaveATP-independentubiquitinligaseactivity.
