Microtubulesarekeycytoskeletalelementsthatarefoundinalleukaryoticcells.Microtubulesfulfilalargerangeofdifferentfunctions,whicharethoughttobecontrolledbythe‘tubulincode’–mechanismtogeneratedistinctmicrotubuleidentities.Onemechanismtolabelspecificmicrotubulesaretubulinposttranslationalmodifications(PTMs),ofwhichalargevarietyexists.Oneofthesemodificationsisglycylation,whichisgeneratedbytheadditionofsecondary(branched)glycinechainstothemain(primary)peptidechainoftheprotein.Thelengthofthesebranchchainscanvaryfromonetomorethan20glycineresidues.GlycylationiscatalysedbytheenzymesTTLL3,TTLL8andTTLL10fromthetubulintyrosineligase-like(TTLL)family.EspeciallyTTLL3andTTLL8areessentialfortheinitiationoftheglycylationbecausethegeneratethenascentglycinechain.
