Transglutaminase(TGase)catalysesthecrosslinkofproteinsbyformingε-(γ-glutamyl)lysineisopeptidebondsandrequiresthebindingofCa2+foritsactivity.Inmammals,eightdistinctTGaseisoenzymeshavebeenidentified.Tissuetransglutaminase(tTGase),alsoknownasTGase2,hasfourdistinctdomains:N-terminalβ-sandwich,catalyticcoreandtwoC-terminalβ-barreldomains.tTGasemayhavearoleincelldeath,cellproliferation,celldifferentiation,andreceptor-mediatedendocytosis.IntheAlzheimer’sdiseasebrain,theelevatedtTGaseactivityismanifestedbypolymerizationofanumberofproteins,includingAβpeptide,β-amyloidprecursorproteinandthetauprotein,withformationofneurofibrillarytangles.Catalyzesthecross-linkingofproteinsandtheconjugationofpolyaminestoproteins.
