Transferrin(Tf)isabloodplasmaproteinforirondelivery.Transferrinfunctionstodeliverirontocellsviaareceptor-mediatedendocytoticprocessaswellastoremovetoxicfreeironfromthebloodandtoprovideanantibacterial,low-ironenvironment.Tfproteincontains679aminoacidresiduesandhasamolecularweightof~79kD,having2specifichighaffinityFe(III)bindingsites.Themoleculeisstabilizedby19intra-chaindisulfidebondsandisprotectedbythreecarbohydratesidechains.Althoughironboundtotransferrinislessthan0.1%(4mg)ofthetotalbodyiron,itisthemostimportantironpool,withthehighestrateofturnover(25mg/24h).WhenaTfproteinloadedwithironencountersatransferrinreceptor(TfR)onthesurfaceofacellitbindstoitandisconsequentlytransportedintothecellbyendosomes.ATP-dependentprotonpumpsthenforceH+ionsintotheendosomesreducingthepHto5.5,thuspromotingironreleaseandconformationalchangeofthereceptorenablingapo-transferrintoremainbound.Thebindingcharacteristicsoftheapo-Tf–TfRcomplexaresuchthattheapo-Tfisreleasedonlyoncethecomplexreachesthecellsurface.CellsexpressinglargeamountsofTfRarecorrespondinglycompetentinsecuringironfromTf.Rapidlyproliferatingcells,asinmalignancy,generallyexpressTfR1abundantly.SeveralstudieshaveshownalinkbetweenTfpolymorphismandsusceptibilitytodisease.Theseincludetherareautosomalrecessivedisorderatransferrinemia,cardiovasculardisease(CVD)andAlzheimer’sdisease.TransferrinsareironbindingtransportproteinswhichcanbindtwoFe3+ionsinassociationwiththebindingofananion,usuallybicarbonate.ItisresponsIBLeforthetransportofironfromsitesofabsorptionandhemedegradationtothoseofstorageandutilization.Serumtransferrinmayalsohaveafurtherroleinstimulatingcellproliferation.
