TLR9(Toll-likeReceptor9)isamemberoftheTLR(Toll-likereceptor)familythatplaysafundamentalroleinpathogenrecognitionandactivationofinnateimmunity.TLR9isexpressedbynumerouscellsoftheimmunesystemsuchasBlymphocytes,monocytes,naturalkiller(NK)cellsandplasmacytoiddendriticcells.Itisexpressedintracellularly,withintheendosomalcompartmentsandfunctionstoalerttheimmunesystemofviralandbacterialinfectionsbybindingtoDNArichinCpGmotifs.Thisnucleotide-sensingTLRisactivatedbyunmethylatedcytidine-phosphate-guanosine(CpG)dinucleotides(alsocalledoligodinucleotides,ODNs),andconsequentlysignalsviaMYD88andTRAF6,leADIngtoNF-κBactivation,cytokinesecretionandtheinflammatoryresponseandeventuallyactivationofaTh1-biasedadaptiveimmuneattack.SpecificTLR9agoNISTshavebeenshowntohavesubstantialantitumoractivity,makingtheTLR9signalingpathwayaninterestingtargetfortherapeuticresearch.
