TLR5(Toll-likeReceptor5)isamemberoftheToll-likereceptor(TLR)familywhichplaysafundamentalroleinpathogenrecognitionandactivationofinnateimmunity.TLR5recognizesbacterialflagellin,aprincipalcomponentofbacterialflagellaandavirulencefactor,viaaconservedregionwithinflagellin,theD1portion/domain.BindingofflagellininducesthedimerizationofTLR5,whichinturnrecruitsMyD88.TherecruitmentofMyD88leadstosubsequentactivationofIRAK4,IRAK1,TRAF6andeventuallyIκBkinases.ActivationofIκBkinasesmobilizesthenuclearfactorNF-κBandstimulatesmanydownstreamgeneexpressions,whichinitiatethecanonicalproinflammatorypathway,includingTNF-αproduction.TLR5mayplayaroleininflammatoryboweldisease(IBD).LowerlevelsofTLR5expressionhavebeenfoundinpatientsexhibitingmoderatetosevereulcerativecolitis(UC).TLR5issuggestedtobepossIBLyinvolvedinHPVinducedinflammationandsubsequentcervicalneoplasiaformation.IthasbeenreportedthatTLR5expressionisdetectedinbothovarianepitheliumandovariancancercelllines,suggestingapossibleroleofTLR5ininflammationinducedovariancanceronset.Mutationsinthisgenehavebeenassociatedwithbothresistanceandsusceptibilitytosystemiclupuserythematosusandsusceptibilitytolegionnairedisease,highlightingtheimportanceofTLR5inmicrobialrecognitionparticularlyatthemucosalsurface.
