TLR4(Toll-likeReceptor4)isamemberoftheToll-likereceptor(TLR)familywhichplaysafundamentalroleinpathogenrecognitionandactivationofinnateimmunity.ItwasthefirstTLRidentifiedTLRinmammalsandmoststudiedmemberoftheTLRfamily.TLR4recognizestheactivelipidAcomponentoflipopolysaccharide(LPS)fromGram-negativebacteriainconjunctionwithLY96(alsoreferredtoMD-2),CD14andLPS-bindingprotein(LBP)andconsequentlymediatestheinnateimmuneresponsetobacteriallipopolysaccharide(LPS).Additionally,itisalsoinvolvedinLPS-independentresponsestriggeredbyendogenousDMAPs,suchasheatshockproteins(HSPs)orHMGB1.TLR4signalsviaMYD88,TIRAPandTRAF6,leADIngtoNF-κBactivation,cytokinesecretionandtheinflammatoryresponseinvariouscelltypes.IncomplexwithTLR6,itwasshowntopromotesterileinflammationinmonocytes/macrophagesinresponsetooxidizedlow-densitylipoprotein(oxLDL)oramyloid-β42.TLR4-mediatedinflammation,triggeredbyexogenousorendogenousligands,isinvolvedinseveralacuteandchronicdiseases,havingapivotalroleasamplifieroftheinflammatoryresponse.ThepotencywithwhichthisTLRactivatesinflammatorypathwaysmakesitanidealtargetfortherapeuticinterventionandadjuvantdevelopment.AbroadrangeofclinicalstudieshaveexaminedtheadjuvantactivityofTLR4activatorsinvaccinesagainstinfectiousandtumorantigens.
