TIE-2actsascell-surfacereceptorforANGPT1,ANGPT2andANGPT4andregulatesangiogenesis,endothelialcellsurvival,proliferation,migration,adhesionandcellspreADIng,reorganizationoftheactinCytoskeleton,butalsomaintenanceofvascularquiescence.Ithasanti-inflammatoryeffectsbypreventingtheleakageofproinflammatoryplasmaproteinsandleukocytesfrombloodvessels.Itisrequiredfornormalangiogenesisandheartdevelopmentduringembryogenesisandforpost-natalhematopoiesis.Afterbirth,itactivatesorinhibitsangiogenesis,dependingonthecontext.Itinhibitsangiogenesisandpromotesvascularstabilityinquiescentvessels,whereendothelialcellshavetightcontacts.Inmigratingendothelialcellsthatlackcell-celladhesions,ANGT1recruitsTEKtocontactswiththeextracellularmatrix,leadingtotheformationoffocaladhesioncomplexes,activationofPTK2/FAKandofthedownstreamkinasesMAPK1/ERK2andMAPK3/ERK1andultimatelytothestimulationofsproutingangiogenesis.
