Thioredoxins(Trx)aresmall,multi-functionalproteinswithoxidoreductaseactivityandareubiquitousinessentiallyalllivingcells.Trxcontainsaredox-activedisulfide/dithiolgroupwithintheconservedCys-Gly-Pro-Cysactivesite.Thetwocysteineresiduesintheconservedactivecenterscanbeoxidizedtoformintramoleculardisulfidebonds(1).ReductionoftheactivesitedisulfideinoxidizedTrxiscatalyzedbyTrxreductasewithNADPHastheelectrondonor.ThereducedTrxisahydrogendonorforribonucleotidereductase,theessentialenzymeforDNAsynthesis,andapotentgeneralproteindisulfidereductasewithnumerousfunctionsingrowthandredoxregulations(2).SpecificproteindisulfidetargetsforreductionbyTrxincludeproteindisulfide–isomerase(PDI)(3)andanumberoftranscriptionfactorssuchasp53(4),NF-κB(5)andAP-1(T1-151).TrxisalsocapableofremovingH2O2,particularlywhenitiscoupledwitheithermethioninesulfoxidereductaseorseveralisoformsofperoxiredoxins(6-7).ParticipatesinvariousredoxreactionsthroughthereversIBLeoxidationofitsactivecenterdithioltoadisulfideandcatalyzesdithiol-disulfideexchangereactions.PlaysaroleinthereversibleS-nitrosylationofcysteineresiduesintargetproteins,andtherebycontributestotheresponsetointracellularnitricoxide.NitrosylatestheactivesiteCysofCASP3inresponsetonitricoxide(NO),andtherebyinhibitscaspase-3activity.ADFaugmentstheexpressionoftheinterleukin-2receptorTAC(IL2R/P55).
