Syk,Spleentyrosinekinase,isexpressedinawiderangeofhematopoieticandnon-hematopoieticcellsandplaysakeyroleinadaptiveimmunesystem,whichistheimmunoreceptor(BcellreceptorandFc-receptors)signalingpathwaysincludingFcγreceptor-mediatedphagocytosis.InhematopoieticcellssuchasBandTlymphocytes,naturalkillercells,mastcells,macrophagesandplatelets,Sykisinvolvedintheproximalsignalingdownstreamofactivatedimmunoreceptors(e.g.BCR,TCR,Fcreceptors)containingimmunoreceptortyrosine-basedactivationmotifs(ITAMs)towhichitbindsusingitstandemSH2domains.ActivatedandautophosphorylatedSykthenphosphorylatesitsspecificsubstratesincludingotherenzymesandadaptorproteins,orchestratingacomplexseriesofcellularresponsessuchascellproliferation,differentiation,survivalandphagocytosis.PositiveeffectorofBCR-stimulatedresponses.CouplestheB-cellantigenreceptor(BCR)tothemobilizationofcalciumioneitherthroughaphosphoinositide3-kinase-dependentpathway,whennotphosphorylatedontyrosinesofthelinkerregion,orthroughaphospholipaseC-γ-dependentpathway,whenphosphorylatedonTyr-348andTyr-352.ThusthedifferentialphosphorylationofSykcandeterminethepathwaybywhichBCRiscoupledtotheregulationofintracellularcalciumion
