Signaltransducerandactivatoroftranscription(STAT),namedaftertheirdualrole,generallymediatecytokine,growthfactorandhormonereceptorsignaltransduction.Inmammals,sevenSTATproteinshavebeenidentified.STAT5hasbeenimplicatedincellularfunctionsofproliferation,differentiationandapoptosiswithrelevancetoprocessesofhematopoiesisandimmunoregulation,reproduction,andlipidmetabolism.TwohighlyhomologousSTAT5isoforms,96kDaSTAT5aand94kDaSTAT5b,areencodedbytwotandemlylinkedgenes.AlthoughbothSTAT5isoformsareroughly95%homologousatthelevelofCDNA,theyexhibitbothredundantandnon-redundantfunctionsinvivo,probablyduetodifferencesintheirtransactivationdomain.AberrantregulationofSTAT5hasbeenobservedinsolidtumorsaswellasinpatientswitheitherchronicoracutemyeloidleukemia.KinaseinhibitorsarecurrentlybeingdevelopedtonegativelyregulateSTAT5activityforclinicalpurposes.Carriesoutadualfunction:signaltransductionandactivationoftranscription.BindstotheGASelementandactivatesPRL-inducedtranscription.
