TheSmadfamilyofproteinsarefunctioninginthetransmissionofextracellularsignalsintheTGF-βsignalingpathway.BindingofaTGF-βsuperfamilyligandstoextracellularreceptorstriggersphosphorylationofSmad2ataSerine-Serine-Methionine-Serine(SSMS)motifatitsC-terminus.PhosphorylatedSmad2isthenabletoformacomplexwithSmad4.Thesecomplexesaccumulateinthecellnucleus,wheretheyaredirectlyparticipatingintheregulationofgeneexpression.Inmammals,eightSmadproteinshavebeenidentifiedtodate.TheSmadfamilyofproteinscanbedividedintothreefunctionalgroups:thereceptor-activatedSmads(R-Smads),commonmediatorSmads(Co-Smads),andtheinhibitorySmads(I-Smads).TheR-SmadsaredirectlyphosphorylatedbytheactivatedtypeIreceptorsontheirC-terminalSer-Ser-X-Ser(SSXS)motifandincludeSmad1,Smad2,Smad3,Smad5,andSmad8.Smad2andSmad3arephosphorylatedinresponsetoTGF-βandactivin,whereasSmad1,Smad5,andSmad8arephosphorylatedinresponsetoBMP(BoneMorphogeneticProtein).ThisC-terminalphosphorylationallowsR-SmadbindingtoCo-Smad,Smad4,andtranslocationtothenucleuswheretheyregulateTGF-βtargetgenes.Smad6andSmad7belongtotheI-SmadwhichbindtothetypeIreceptororSmad4andblocktheirinteractionwithR-Smads.TheSmadssharesequencesimilarities,mostnotablyintheN-terminalandcarboxy-terminalregions,referredtoastheMH1(MadHomology1)andMH2domainsrespectively.Smad2andSmad3have66%aminoacidsequenceidentitybetweentheirMH1domainsand96%aminoacidsequenceidentitybetweentheirMH2domains.TranscriptionalmodulatoractivatedbyTGF-βandactivintype1receptorkinase.SMAD2isareceptor-regulatedSMAD(R-SMAD).Mayactasatumorsuppressorincolorectalcarcinoma.
