SHP2isatyrosinephosphatasecontainingtwotandemSH2(srchomology2)domains.Proteintyrosinephosphatases(PTPs)areagroupofenzymesthatremovephosphategroupsfromphosphorylatedtyrosineresiduesonproteins.Togetherwithtyrosinekinases,PTPsregulatethephosphorylationstateofmanyimportantsignalingmolecules.SHP1andSHP2representasubfamilyofnon-transmembranePTPsthatcontaintwoSH2domainsfollowedbyaPTPdomain.TyrosinephosphorylationofSHP2isrequiredfornormalERKactivationinresponsetosomegrowthfactors.Intheinactivestate,theN-terminalSH2domainbindsthePTPdomainandblocksaccessofpotentialsubstratestotheactivesite.Thisauto-inhibitionisrelievedbythebindingofphosphopeptidestoSH2domain,resultinginactivationofphostphataseactivity.Noonansyndromecharacterizedbyanabnormalface,shortstatureandcardiacabnormalitiesisduetothemutationsofN-terminalSH2domainorPTPdomain.SHP2mutationsmightalsocausesomeleukemia,andcouldbeimportantinpathogenesisbyHelicobacterpylori,themajorcauseofgastriculcerandcarcinoma.Actsdownstreamofvariousreceptorandcytoplasmicproteintyrosinekinasestoparticipateinthesignaltransductionfromthecellsurfacetothenucleus.
