SHP1andSHP2representasubfamilyofnon-transmembraneprotein-tyrosinephosphatases(PTPs)thatcontaintwotandemSH2(srchomology2)domains.SHPshavetwoN-terminalSH2domains(N-SH2andC-SH2),aclassicPTPdomainandaC-terminaltailharboringtwotyrosylphosphorylationsiteswhicharephosphorylateddifferentiallybyreceptorandnon-receptorprotein-tyrosinekinases(PTKs).WhereasSHP2isexpressedubiquitously,SHP1isprimarilyexpressedinhematopoieticcellsandbehavesasakeyregulatorcontrollingintracellularphosphotyrosinelevelsinlymphocytes.SH2domains(particularlytheN-SH2)alsoregulatePTPactivity.BasalSHPactivityislow,butadditionofaphosphotyrosylpeptidethatbindstheN-SH2(Tyr–Ppeptideligand)markedlystimulatescatalysis.SHP1isimplicatedinsignalingfromreceptortyrosinekinases(RTKs),cytokinereceptors,chemokinereceptorsandintegrins.SHP1-deficientbonemarrowmacrophagesarehyper-adherenttoβ1-andβ2-integrinligands.DecreasedSHP1levelcausesabnormalT-lymphocyteproliferationandinducesvarioustypesofleukemias.IntroductionoftheSHP1genebackintoaleukemiacelllineandaprostatecancercelllinedemonstratedthetumorsuppressorfunctionofSHP1.Playsakeyroleinhematopoiesis.ThisPTPaseactivitymaydirectlylinkgrowthfactorreceptorsandothersignalingproteinsthroughprotein-tyrosinephosphorylation.TheSH2regionsmayinteractwithothercellularcomponentstomodulateitsownphosphataseactivityagainstinteractingsubstrates.TogetherwithMTUS1,inducesUBE2V2expressionuponangiotensinIIstimulation.
