Thep90ribosomalS6kinases(RSKs)compriseafamilyofserine/threoninekinasesthatlieattheterminusoftheERKpathway.Inhumans,theRSKfamilyconsistsoffourisoforms(RSK1to-4).RSKfamilymembersareunusualamongserine/threoninekinasesinthattheycontaintwodistinctkinasedomains,bothofwhicharecatalyticallyfunctional.Theseskinasedimainsareactivatedinasequentialmannerbyaseriesofphosphorylations.RSKregulatesgeneexpressionviaassociationandphosphorylationoftranscriptionalregulatorsincludingc-Fos,estrogenreceptor,NFκB/IκBα,cAMP-responseelement-bindingprotein(CREB).ERKactivatestheC-terminalkinaseofRSK,leADIngtoactivationoftheN-terminalkinase.MembersoftheRSKfamilyarepresentinthecytoplasmaswellasthenucleus.AdditionofgrowthfactortothecellsresultsintheactivationofbothcytosolicandnuclearRSKandthetranslocationofthecytosolicRSKintothenucleusuponactivation.TheactivationandnucleartranslocationofRSKresultinphosphorylationandactivationoftranscriptionfactors.Serine/threoninekinasethatmayplayaroleinmediatingthegrowth-factorandstressinducedactivationofthetranscriptionfactorCREB.
