Receptorinteractingprotein(RIP)kinasesconstituteafamilyofsevenmembers,allofwhichcontainakinasedomain(KD).Theyarecrucialregulatorsofcellsurvivalanddeath.RIP1,thefirstmemberofthefamily,ishighlyconservedinvertebrateevolution.RIP1andRIP2bearaC-terminaldomainbelongingtothedeathdomain(DD)andacaspaserecruitmentdomain(CARD),allowingrecruitmenttolargeproteincomplexesinitiatingdifferentsignalingpathways.HumanRIP1is68%identicaltomouseRIP1,andthegreatestsimilarityisintheDD.RIP1-DDisimportantforbindingtootherdeathreceptors,suchastumornecrosisfactor(TNF)-R1,TRAIL-R1andTRAIL-R2,andtoDD-containingadaptorproteinssuchasTNF-receptor-associateddeathdomain(TRADD)andFADD.RIP1isconstitutivelyexpressedinmanytissues.However,TNFtreatmentandT-cellactivationcanalsoinduceRIP1expression.Full-lengthRIP1isimportantforsignalingtoNF-B,MAPKsandnecrosis,whereascaspase-8generatedC-terminalRIP1cleavagefragmentpromotesapoptosisasapositivefeedbackloop.RIP1thusintegratesseveraldifferentupstreamsignalstoinitiatealimitednumberofcellularresponses.RIP1isacrucialadaptorkinaseinseveralofthestress-inducedsignalingpathways.Theimbalancebetweencellsurvivalanddeath,akeyfeatureofmanyneoplastic,degenerative,inflammatory,andautoiummunediseases,maybecausedbyaberrantreactiveoxygenspecies(ROS)turnover,whichcanberegulatedbydeathreceptors(DRs)andtheireffectorpathways.InvolvedinthetransductionofTNFsignaling.TNFstimulationinducesbindingofRIPK1-TRADD-TRAF2-TRAF5complextoTNFR1.TRAFproteinsthencatalyzethe"Lys-63"-linkedpolyubiquitinationofRIPK1,inducingRIPK1associationwiththeIKKcomplex,whichissubsequentlyactivated,leADIngultimatelytotheactivationofNF-κ-B.TheinflammatoryresponseiskepttransientbytheactionoftheNF-κ-BtargetgeneproductTNFAIP3.TNFAIP3isrecruitedtoRIPK1withinacomplexcomprisingalsoRNF11,ITCHandTAX1BP1.TNFAIP3deubiquitinates"Lys-63"-polyubiquitinchainsonRIPK1andcatalyzestheformationof"Lys-48"-polyubiquitinchains.ThisleadstoRIPK1proteosomaldegradationandconsequentlytotheterminationoftheTNF-orLPS-mediatedactivationofNF-κ-B.
