TheRassuperfamilyofsmallGTPasesincludestheRas,Rho,Arf,Rab,andRanfamilies.Ras-regulatedsignalpathwayscontrolsuchasactincytoskeletalintegrity,proliferation,differentiation,celladhesion,apoptosis,andcellmigration.TheseGTPasesfunctionasmolecularswitchesthatcontroleukaryoticcellfunctionbycyclingbetweentwointerconvertIBLeforms,aGDP-boundinactiveformandaGTP-boundactiveform.ThebindingofGTPleadstoaconformationalchangeinthedownstreameffector-bindingdomainoftheG-protein.ThesmallGTPasesaremonomericG-proteinswithmolecularmassesovertherange20–30kDa.Rasisattachedtothecellmembranebyprenylation.MutationsintheRasfamilyofproto-oncogenes(comprisingH-Ras,N-RasandK-Ras)areverycommon,beingfoundin20%to30%ofallhumantumours.ThefrequencyofRASmutationsisamongthehighestforanygeneinhumancancer.K-rasmutationsoccurfrequentlyinnon-small-celllung,colorectal,andpancreaticcarcinomas;H-rasmutationsarecommoninbladder,kidney,andthyroidcarcinomas;N-rasmutationsarefoundinmelanoma,hepatocellularcarcinoma,andhematologicmalignancies.RegulatestheorganizationoftheactinCytoskeleton.
