Receptorproteintyrosinephosphataserho(RPTPρ/PTPRT)isatransmembraneproteinthatishighlyexpressedinthedevelopingandadultcentralnervoussystem.ItisamemberoftheRPTPR2Bsubfamily,whichincludesPTPκ,PTPμandPCP-2.TheR2Bphosphatasesareknowntointeractwithmembersofthecadherin/catenincomplex.ThesefourRPTPssharethesamedomainstructure:anextracelluardomain,ajuxtamembraneregion,andtwophosphatasedomains.TheextracellulardomainofPTPRmediatescell-cellaggregationandthatmutationalinactivationofthisphosphatasecouldpromotetumormigrationandmetastasis.PTPRTisthemostfrequentlymutatedPTPinhumancancers.STAT3isasubstrateofPTPRT.Phosphorylationofatyrosineat705isessentialforthefunctionofSTAT3,andPTPRTspecificallydephosphorylatedSTAT3atthisposition.MaybeinvolvedinbothsignaltransductionandcellularadhesionintheCNS.
