Receptor-typetyrosine-proteinphosphataseeta(alsoknownasDEP1;SCC1;CD148;HPTPeta;R-PTP-ETA)isanenzymethatinhumansisencodedbythePTPRJgene[1-3].Theproteinencodedbythisgeneisamemberoftheproteintyrosinephosphatase(PTP)family.PTPsareknowntobesignalingmoleculesthatregulateavarietyofcellularprocessesincludingcellgrowth,differentiation,mitoticcycle,andoncogenictransformation.ThisPTPpossessesanextracellularregioncontainingfivefibronectintypeIIIrepeats,asingletransmembraneregion,andasingleintracytoplasmiccatalyticdomain,andthusrepresentsareceptor-typePTP.ThisPTPispresentinallhematopoieticlineages,andwasshowntonegativelyregulateTcellreceptorsignalingpossIBLythroughinterferingwiththephosphorylationofPhospholipaseCγ1(PLCG1)andLinkerforActivationofTCells(LAT).ThisPTPwasalsofoundtodephosphorylatePDGFβreceptor,andmaybeinvolvedinUV-inducedsignaltransduction[3].PTPRJhasbeenshowntointeractwithCTNND1[4].TyrosinephosphatasewhichdephosphorylatesorcontributestothedephosphorylationofCTNND1,FLT3,PDGFRB,MET,RET(variantMEN2A),KDR,LYN,SRC,MAPK1,MAPK3,EGFR,TJP1,OCLN,PIK3R1andPIK3R2.Playsaroleincelladhesion,migration,proliferationanddifferentiation.Involvedinvasculardevelopment.RegulatorofmacrophageadhesionandspreADIng.Positivelyaffectscell-matrixadhesion.Positiveregulatorofplateletactivationandthrombosis.Negativeregulatorofcellproliferation.NegativeregulatorofPDGF-stimulatedcellmigration;throughdephosphorylationofPDGFR.Positiveregulatorofendothelialcellsurvival,aswellasofVEGF-inducedSRCandAKTactivation;throughKDRdephosphorylation.NegativeregulatorofEGFRsignalingpathway;throughEGFRdephosphorylation.Enhancesthebarrierfunctionofepithelialjunctionsduringreassembly.NegativelyregulatesT-cellreceptor(TCR)signaling.UponT-cellTCRactivation,itisup-regulatedandexcludedfromtheimmunologicalsynapses,whileuponT-cell-antigenpresentingcells(APC)disengagement,itisnolongerexcludedandcandephosphorylatePLCG1andLATtodown-regulateprolongationofsignaling.
