Focaladhesionkinasesubfamilyconsistsofthenon-receptorproline-richproteintyrosinekinases(PTKs).Twomembersofthefamilyarefocaladhesionkinase(FAK)andproline-richtyrosinekinase2(PYK2).Thesetwokinaseshavemolecularmassbetween110-125kDaandarecloselyrelatedintheirstructure.Thepresenceoftwoproline-richmotifswithintheC-terminaldomainsisconserved.Pyk2isexpressedprimarilyinbrainandhematopoieticcellsandbecomesactivatedinresponsetostimulationthroughnumerousreceptors,includingintegrins,chemokinereceptors,andantigenreceptors.Itisalsoexpressedinbothinbone-formingosteoblastsandbone-resorbingosteoclasts,andthoughttohavepositiveroleinosteoblastmaturationandboneresorption.PhosphorylationofPyk2leadstotherecruitmentofSrcfamilykinasesandtheactivationofErks.Pyk2alsointeractswithandphosphorylatesthefocaladhesion-relatedprotein,paxillin,andothercytoskeletalproteins,suggestingitspivotalroleinvariouscellularevents.Involvedincalciuminducedregulationofionchannelandactivationofthemapkinasesignalingpathway.Mayrepresentanimportantsignalingintermediatebetweenneuropeptideactivatedreceptorsorneurotransmittersthatincreasecalciumfluxandthedownstreamsignalsthatregulateneuronalactivity.InteractswiththeSH2domainofGrb2.Mayphosphorylatethevoltage-gatedpotassiumchannelproteinKv1.2.Itsactivationishighlycorrelatedwiththestimulationofc-JunN-terminalkinaseactivity.Involvedinosmoticstress-dependentSNCA"Tyr-125"phosphorylation.
