ProthrombinprecursorissynthesizedinliverasaprecursorofprothrombincontainingasignalsequenceandapropeptidedomainattheN-terminus.Beforeitssecretionintoplasma,thisprecursorundergoesseveralposttranslationalmodifications,includingremovalofsignalsequenceandpropeptidedomains,glycosylation,andγ-glutamyl-carboxylationreaction.Prothrombin,cleavedbytheprothrombinaseenzymecomplexthatconsistsofserineproteasefactorXa,cofactorVa,phospholipidsandcalcium,isconvertedtothrombinwhichconvertsfibrinogenintofibrinwhichinturnstrengthensaprotectiveclot.Activationofprothrombiniscrucialinphysiologicalandpathologicalcoagulation.Variousrarediseasesinvolvingprothrombin(e.g.hypoprothrombinemia)havebeendescribed.Thrombin,whichcleavesbondsafterArgandLys,convertsfibrinogentofibrinandactivatesfactorsV,VII,VIII,XIII,and,incomplexwiththrombomodulin,proteinC.Functionsinbloodhomeostasis,inflammationandwoundhealing.
