Profilin(PFN1)isaubiquitoussmall(12-15kDa)phosphoinositideandpoly-L-prolinebindingproteinthatplaysaroleinsignaltransductionpathwaysandactinfilamentdynamics.Therearetwomammalianprofilinswithsimilarbiochemicalproperties.WhereasprofilinIappearstobehighlyexpressedinmosttissuesexceptforskeletalmuscle,profilinIIispredominantlyexpressedinbrainandatlowerlevelsalsoinskeletalmuscle,uterusandkidney.ProfilinisamainlycytosolicproteinwithhigherconcentrationsindynamicmembraneareasliketheleADIngedgeandrufflingmembranes.ProfilinbindingtoPIP2interfereswithPIP2hydrolysisbysolublephospholipaseC-γ,aninhibitionthatcanbeovercomebytyrosinephosphorylationofPLC-γ.Besidesactinmonomersequestrationandstimulationofactinnucleotideexchange,profilincanalsopromotecellularactinfilamentgrowth.Profilinisinvolvedintheactindependentintracellularmotilityofcytopathogenicbacteria,theregulationofcelladhesionandpossIBLyalsoinlinkingtheactinCytoskeletonandendocytosis.ProfilinhasbeenfoundtoassociatewithdefinedcomplexescontainingproteinssuchasArp2/3ortheRho/RacpathwaysconstituentsROCK-IIandHEM2/NAP1.DefectsinPFN1arethecauseofamyotrophiclateralsclerosis18(ALS18).
