ProteinPhosphatase2A(PP2A)isoneofthemajorSer/Thrphosphatasesimplicatedintheregulationofmanycellularprocessesincludingregulationofdifferentsignaltransductionpathways,cellcycleprogression,DNAreplication,genetranscriptionandproteintranslation.Thecorestructurecomprisesa36kDacatalyticsubunit(PP2AC)anda65kDaregulatorysubunit(PR65orAsubunit).EachPP2Asubunithasatleasttwoisoformsandthecatalyticsubunit,presentintheαandβisoforms,share97%homology.Thedifferentialassociationofallthesesubunitsgivesrisetoanextensivesubsetofoligomericholoenzymes.ItiswidelythoughtthatPP2Aexercisesregulatoryflexibilityanddifferentialsubstratespecificitythroughthespecificassociationofthecoredimer(PP2AD)withoneofthethreeregulatoryBsubunits.Moreover,PP2Ainteractswithastillgrowingnumberofcellularandviralproteinsandisregulatedbyposttranslationalmodifications.PP2AcanmodulatetheactivityofphosphorylaseBkinasecaseinkinase2,mitogen-stimulatedS6kinase,andMAP-2kinase.CandephosphorylateSV40largeTantigenandp53.DephosphorylatesSV40largeTantigen,preferentiallyonserineresidues120,123,677,andperhaps679.TheCsubunitwasmostactive,followedbytheACform,whichwasmoreactivethantheABCform,andactivityofallthreeformswasstronglystimulatedbymanganese,andtoalesserextentbymagnesium.DephosphorylationbytheACform,butnotCorABCformisinhibitedbysmallTantigen.
