Paraoxonasesareagroupofenzymesinvlovedinthehydrolysisoforganophosphates.Paraoxon,anorganophosphate,isanacetylcholinesteraseinhibitorandtheactivemetaboliteoftheinsecticideparathion.Besidesatoxicologicalrole,clinicalinteresthasfocusedonaprotectiveroleinvasculardisease.Paraoxonase-1(PON1)isacalcium-dependentesterhydrolasethatistightlyassociatedwithapoA-IinHDL.PON1preventstheoxidationofLDL,therebytheformationofatherogenicoxidised-LDL.PolymorphismsofthehumanPON1genearecorrelatedwithcoronaryarterydisease,indicatingageneticassociationbetweenPON1andcoronaryarterydisease.PON1activityislowerinsubjectspronetodevelopmentofatherosclerosissuchasintype1andtype2diabetes,familialhypercholesterolemia,andrenaldisease,indicatingtheinvolvementofPON1inatherosclerosisdevelopment.PON2isaubiquitouslyexpressedintracellularproteinthatcanprotectcellsagainstoxidativedamage.PON3issimilartoPON1inactivitybutdiffersfromitinsubstratespecificity.PON1andPON3areimplicatedinloweringtheriskofdevelopingcoronaryarterydiseaseandatherosclerosis.HumanPON3proteinsharesthe3conservedcysteineresiduesidentifiedinPON1,suggestingtheirimportanceofinvivoactivities.PON1-basedcatalyticscavengerscanbeawaytosafeandeffectiveorganophosphateintoxication.Hydrolyzesthetoxicmetabolitesofavarietyoforganophosphorusinsecticides.Capableofhydrolyzingabroadspectrumoforganophosphatesubstratesandanumberofaromaticcarboxylicacidesters.Hasantioxidantactivity.Isnotassociatedwithhighdensitylipoprotein.PreventsLDLlipidperoxidation,reversestheoxidationofmildlyoxidizedLDL,andinhibitstheabilityofMM-LDLtoinducemonocytechemotaxis.
