MicrotubulesarekeyelementsoftheeukaryoticCytoskeletonthatdynamicallyassemblefromheterodimersofα-andβ-tubulin.Twodifferentmechanismscangeneratemicrotubulediversity:theexpressionofdifferentα-andβ-tubulingenes,referredtoastubulinisotypes,andthegenerationofposttranslationalmodifications(PTMs)onα-andβ-tubulin.TubulinPTMsincludethewell-knownacetylationorphosphorylation,andothersthathavesofarmostlybeenfoundontubulin,detyrosination/tyrosination,polyglutamylationandpolyglycylation.ThesePTMsmighthaveevolvedtospecificallyregulatetubulinandmicrotubulefunctions.PolyglutamylationisaPTMthatoccurswhensecondaryglutamatesidechainsareformedonγ-carboxylgroupsofglutamateresiduesinaprotein.EnzymescatalyzingpolyglutamylationbelongtotheTTL-like(TTLL;Tubulintyrosineligase-like)familyofglutamylases.Deglutamylases,theenzymesthatreversepolyglutamylation,wereidentifiedwithinanovelfamilyofCCPs(cytosoliccarboxypeptidase).Subtledifferencesinpolyglutamylationcanbeseenondiversemicrotubulesindifferentcelltypes.Thefunctionsofthesemodificationsremaintobestudied.However,itswidedistributionstrengthenstheideathatitcouldbeinvolvedinfine-tuningarangeofmicrotubulefunctions.PolyElabelscentriolesastheymature,suchthattwofociarepresentthroughoutthecellcycle.
