Polo-likekinases(Plks)areimportantregulatorsofthecellcycle.Plk1,themoststudiedPlk,hasbeenimplicatedinregulatingcentrosomematuration,mitoticentry,sisterchromatidcohesion,theanaphase-promotingcomplex/cyclosome(APC/C),andcytokinesis.Sisterchromatidseparationandthesubsequentformationoftwogeneticallyidenticaldaughtercellsdependonthesymmetricalattachmentofallchromosomestothemitoticspindle,aprocesscalledchromosomebiorientation.Severalmitosis-specificproteinkinaseshavebeenimplicatedinbipolarspindleassemblyandchromosomebiorientation.Forexample,Cdk1(cyclin-dependentkinase1)isknowntophosphorylateEg5,akinesinthatisrequiredforthemigrationofcentrosomesandisthusneededfortheformationofbipolarspindles.Attheonsetofmitosis,PlkscontributetotheactivationofCdk1-cyclinB,andtheyarealsorequiredfortheinactivationofCdk1andexitfrommitosis.PlksareimportantregulatorsoftheAPC/C,akeycomponentoftheubiquitin-dependentproteolyticdegradationpathway.Serine/threonine-proteinkinasethatperformsseveralimportantfunctionsthroughoutMphaseofthecellcycle,includingtheregulationofcentrosomematurationandspindleassembly,theremovalofcohesinsfromchromosomearms,theinactivationofAPC/Cinhibitors,andtheregulationofmitoticexitandcytokinesis.
