ProteinkinaseC(PKC)isafamilyofserine-threoninekinasesthatregulateabroadspectrumofcellularfunctions.Thefamilyiscomposedofninegenesthatexpressstructurallyrelatedphospholipid-dependentkinaseswithdistinctmeansofregulationandtissuedistribution.BasedontheirstructuresandsensitivitiestoCa2+anddiacylglycerol(DAG),theyhavebeenclassifiedintoconventionalPKCs(α,β,andγ),novelPKCs(δ,ε,η,andθ),andatypicalPKCs(ζandλ/ι).Anovelserine-threoninekinaseoftheproteinkinaseC(PKC)familyhasbeendescribedanddesignatedasPKCν(PKCν)whichhastwoputativediacylglycerolbindingC1domains.PKCνisabundantlyexpressedinhumanβ-cells,anditisadownstreameffectorforBCR(B-cellantigenreceptor)-mediatedDAGproduction.TheclosesthomologuesofPKCνarePKD1/PKCμandPKD2,andtogetherthesethreekinasesformadistinctproteinkinasesubfamily.TheyshareapredictedtertiarystructurethatincludestwoC1domainscontainedintheiramino-terminalhalves,asinglecentralpHdomain,andcloselyhomologouskinasedomainsintheirCOOH-terminalhalves.Convertstransientdiacylglycerol(DAG)signalsintoprolongedphysiologicaleffects,downstreamofPKC.Involvedinresistancetooxidativestress.
