ProteinkinaseC(PKC)isafamilyofserine-threoninekinasesthatregulateabroadspectrumofcellularfunctions.Thefamilyiscomposedofninegenesthatexpressstructurallyrelatedphospholipid-dependentkinaseswithdistinctmeansofregulationandtissuedistribution.BasedontheirstructuresandsensitivitiestoCa2+anddiacylglycerol(DAG),theyhavebeenclassifiedintoconventionalPKCs(α,β,andγ),novelPKCs(δ,ε,η,andθ),andatypicalPKCs(ζandλ/ι).PKCγisamemberofthecPKCsubfamilywhichisactivatedbyCa2+anddiacylglycerolinthepresenceofphosphatidylserine.Physiologically,PKCγisactivatedbyamechanismcoupledwithreceptor-mediatedbreakdownofinositolphospholipidasothercPKCisotypes.PKCγisexpressedsolelyinthebrainandspinalcordanditslocalizationisrestrictedtoneurons.Withinthebrain,PKCγisthemostabundantinthecerebellum,hippocampusandcerebralcortex,wheretheexistenceofneuronalplasticityhasbeendemonstrated.Thisisacalcium-activated,phospholipid-dependent,serine-andthreonine-specificenzyme.PKCisactivatedbydiacylglycerolwhichinturnphosphorylatesarangeofcellularproteins.PKCalsoservesasthereceptorforphorbolesters,aclassoftumorpromoters.
