ProteinkinaseC(PKC)isafamilyofserine-threoninekinasesthatregulateabroadspectrumofcellularfunctionssuchascellmigrationandpolarity,proliferation,differentiation,andcelldeath.Thefamilyiscomposedofseveralgenesthatexpressstructurallyrelatedphospholipid-dependentkinaseswithdistinctmeansofregulationandtissuedistribution.BasedontheirstructuresandsensitivitiestoCa2+anddiacylglycerol(DAG),theyhavebeenclassifiedintoconventionalPKCs(α,β,andγ),novelPKCs(δ,ε,η,andθ),andatypicalPKCs(ζandλ/ι).PKCsshareastructuralbackbone,mainlyconsistingofaregulatorydomainattheN-terminusandacatalyticdomainattheC-terminus.Allfamilymembersrequirephosphatidylserine,acomponentofthephospholipidbilayer,fortheiractivation.SomePKCs(PKCα,δ,andζ)arewidelyexpressedinalltissues,butotherisoformsareexpressedinatissue-specificmanner.PKCγ,forexample,islargelyconfinedtobrainandneuronaltissue,PKCιismainlyexpressedintestisandinsulinsecretingcells,andPKCθismainlyexpressedinskeletalmuscle.PKCε(PKCε)isacalcium-independentandphorbolester/diacylglycerol-sensitiveserine/threoninekinase.PKCεistheonlyPKCisozymethathasbeenshowntobehaveasanoncoprotein.ConstitutiveactivationofPKCεinasmallcelllungcancer(SCLC)celllineandtheoverexpressionofPKCεincolonicepithelialcellshavebeenreported.Thisiscalcium-independent,phospholipid-dependent,serine-andthreonine-specificenzyme.PKCisactivatedbydiacylglycerolwhichinturnphosphorylatesarangeofcellularproteins.PKCalsoservesasthereceptorforphorbolesters,aclassoftumorpromoters.
